Let’s say I have 3 subs, 4 fighters, and 3 bombers attacking a sub, 3 destroyers, and 3 fighters. Since I’m attacking BUT do not have a destroyer, does it mean the defending player CANNOT use his one sub as a casualty?

If the owner of the defending Sub decided to stay and not submerge it, it can be hit by your attacking Subs. But only by them. Not any of the Air units.
